Peptide degradation is rarely dramatic. It's usually a slow accumulation of oxidation, deamidation, and aggregation that quietly erodes an assay's reliability before anyone notices. Semaglutide is reasonably robust as lyophilized peptides go, but its stability still depends heavily on how it's stored before and after reconstitution.
The lyophilized vial
In its freeze-dried state, semaglutide is comparatively stable because the absence of bulk water limits the hydrolytic and enzymatic pathways that drive peptide breakdown. Sealed vials are typically kept refrigerated at 2–8 °C for shorter-term storage, or frozen at −20 °C or below for longer intervals before use. Heat, humidity, and direct light are the three variables most consistently associated with accelerated loss of potency in stored peptide powders, so vials are generally kept in their original packaging, away from windows and heat sources, until they're needed.
What changes after reconstitution
Once dissolved into a liquid research stock, semaglutide is considerably more exposed to degradation pathways: hydrolysis, oxidation of susceptible residues, and physical aggregation all proceed faster in solution than in the dry state. In transit
Stability considerations don't start at the bench — they start in the shipping box. Lyophilized peptides are generally tolerant of brief excursions above refrigeration temperature during transit, since the dry state buffers against short-term thermal stress far better than a reconstituted solution would. Even so, prolonged exposure to heat during shipping (for example, a package left in a hot vehicle or mailbox for an extended period) can measurably shorten a lot's effective shelf life, which is one reason researchers note the condition of packaging on arrival and store material promptly rather than leaving it at room temperature.
Practical handling notes
- Keep reconstituted solution refrigerated and shielded from light between uses.
- Aliquot into single-use volumes where practical to avoid repeated warming of the full stock.
- Minimize freeze-thaw cycling, which promotes aggregation in many peptide solutions.
- Inspect for cloudiness, particulates, or discoloration before use, and discard visibly compromised solution.
- Label aliquots with reconstitution date so degradation trends can be tracked across an experiment.
